Dopamine transporter (DAT) in the presynaptic neuron modulates the uptake of dopamine, regulating its concentration in central nervous system. [99mTc]TRODAT-1 is a radiopharmaceutical used to investigate dopaminergic neurotransmission showing DAT density. Radioligands allow in vivo evaluation of receptor density, measured as binding potential (BP). The objective of this study was to evaluate DAT density in a sample of brazilian healthy individuals throughout the [99mTc]TRODAT-1 SPECT image analysis. Seventy-three images of healthy individuals (40 men, 33 women), aged from 18 to 62 yo, were selected. Filtered back projection method of reconstruction, Butterworth filter and Chang attenuation correction were applied. Semi-quantitative evaluation was performed in transaxial slices (8 mm) making regions of interest in striatal region (STR) and using the occipital region (OCC) as non-specific DAT-BP (DAT-BP=STR-OCC/OCC). Data showed that DAT-BP decrease in healthy individuals from 30 years of age reaching approximately 45% after 60 years of age. We observed that this decrease per age group is similar in the right (R2linear= 0.502) and left striatum (R2linear=0.456). Women presented higher DATBP than men (womenR2linear=0.555; menR2linear= 0.432), but showed a decrease per age group equal to men. Our study showed important data on healthy subjects DAT-BP values, which will allow us to continue the standardization of DAT in the investigation of dopaminergic neurotransmission.
